The Scary Side Effect of Recently Approved Weight-loss Pills ‘Must Be Considered’ Before Taking Them

Recent advancements in weight loss drugs have brought GLP-1 agonists into the limelight. While they hold promise, their gastrointestinal (GI) implications remain ambiguous.

A groundbreaking study in JAMA has delved deep into the potential GI issues stemming from GLP-1 agonist usage.

GLP-1, a gut hormone triggered by food consumption, interacts with its receptors found throughout our body. The result? A reduced appetite and enhanced glucose absorption into muscle and fat cells leading to lower blood sugar.

But there’s a catch. Natural GLP-1 agonists don’t last long, prompting the creation of synthetic versions like semaglutide and liraglutide. Approved for both type 2 diabetes and weight loss (with the right lifestyle adjustments), these drugs aren’t without controversy. Evidence suggests they might trigger more GI disturbances, albeit typically mild.

Diabetic patients, already predisposed to GI challenges such as pancreatitis, could face unique risks when on these drugs compared to their non-diabetic counterparts.

Unpacking the Study

An extensive analysis was done on data from the PharMetrics Plus database, covering an impressive 90% of all US outpatient prescriptions and doctor diagnoses. Participants were either on semaglutide, liraglutide, or buproprion-naltrexone (a different weight loss drug).

While demographic differences were noted across the drug users, the pivotal observation lay in the higher rates of specific GI issues among GLP-1 agonist users compared to the buproprion-naltrexone group. This encompassed issues like gallbladder challenges, pancreatitis, bowel blockages, and delayed stomach emptying.

A marked elevation in pancreatitis risk stood out for those on GLP-1 agonists, even after accounting for factors like smoking or recent surgeries.

Interestingly, patient obesity history, when taken out of the equation, only slightly modified the findings. Body weight measurements also didn’t seem to affect the outcomes.

Deciphering the Findings

There’s a clear rise in GI risks such as delayed stomach emptying, pancreatitis, and intestinal blockages following GLP-1 agonist administration. However, the biliary disease risk remained unchanged.

The authors conclude, “Given the wide use of these drugs, these adverse events, although rare, must be considered by patients who are contemplating using the drugs for weight loss because the risk-benefit calculus for this group might differ from that of those who use them for diabetes.”
